Krystal Biotech, Inc. (KRYS) announced positive preliminary clinical data for KB113, an investigational immunotherapy designed to treat basal cell carcinomas (BCCs) in patients with Gorlin syndrome. Krystal shares rose 2.97%.
Gorlin syndrome is a rare genetic disorder that significantly increases the risk of developing multiple basal cell carcinomas, often requiring repeated surgeries that can cause scarring, tissue damage and long-term effects on quality of life.
KB113 is an investigational intratumoral immunotherapy designed to promote localized expression of interleukin-2 (IL-2) and interleukin-12 (IL-12) within tumors, potentially stimulating immune-mediated cancer cell clearance.
In a pilot study involving three adult patients with Gorlin syndrome, more than 80% of the assessed BCC lesions showed clinical regression following treatment. Responses were observed in both injected and non-injected lesions, suggesting a potential systemic immune response known as an abscopal effect.
Biopsies of one injected lesion from each patient confirmed complete tumor clearance, with no residual tumor tissue detected. The study evaluated patients over a 14-week treatment period, followed by a final assessment 30 days after the last dose.
KB113 was well tolerated, with all reported adverse events mild to moderate, transient and resolved during the study.
Krystal said it has initiated discussions with the U.S. Food and Drug Administration regarding the further clinical development of KB113 for Gorlin syndrome. The company plans to provide an update on its development strategy following alignment with the FDA.
The findings will be presented by study investigator Dr. Shireen Guide at the 14th Pediatric Dermatology Research Alliance Annual Conference, held October 15-17, 2026, in Alexandria, Virginia.
