AstraZeneca(AZN.US)

  • AVANZAR(Datroway一线肺癌)等近期催化剂与FOMC宏观波动可能掩盖或放大个股反应

AstraZeneca’s newly approved pill fails frontline breast cancer study
Etcamah is the second oral “SERD” to recently fall short in first-line breast cancer, following a similar setback from Roche earlier this year.

A newly approved drug from AstraZeneca has failed a major trial that could have supported broader use of the therapy in frontline breast cancer.
According to AstraZeneca, a combination of its therapy Etcamah and a commonly used medication
failed to significantly delay disease progression
compared to that standard medicine and hormone therapy in people newly diagnosed with advanced ER-positive, HER2-negative breast cancer. The company didn’t provide specifics in its statement Friday, but said that treatment was associated with a “numerical improvement” in progression-free survival that didn’t reach statistical significance.
Detailed data will be shared “in due course,” AstraZeneca said.
Etcamah is part of a class of oral medicines called selective estrogen receptor degraders, or SERDs. These treatments are being developed as alternatives to the decades-old injectable breast cancer therapy fulvestrant. A few have recently gotten to market, and another could be cleared later this year.
Drug companies, however, have struggled to prove that oral SERDs will become the broadly used medicines they’d originally anticipated. The benefits have been most pronounced among a subset of breast cancer patients with “ESR1” mutations that help tumors develop resistance to treatment.
AstraZeneca has been hoping for better with Etcamah. The drug won an
accelerated approval
earlier this month in a novel setting, where treatment is recommended as a part of first-line care once an ESR1 mutation is detected by an FDA-approved test. That clearance supported expectations from AstraZeneca executives that the therapy might eventually rake in
$5 billion annually
, making Etcamah a key part of the company’s plans to
reach $80 billion in yearly sales
by 2030.
The study AstraZeneca reported on Friday, “SERENA-4,” was meant to be another step forward in that plan, as it evaluated Etcamah as part of a standard frontline regimen. But expectations dimmed after another oral SERD
from Roche called giredestrant failed a study in a similar setting
. SERENA-4 was a “high-risk, high-reward” study, wrote Leerink Partners analyst Andrew Berens. He’d given the study a 20% probability of succeeding.
AstraZeneca is awaiting the results of another study important to its future. Called “AVANZAR,” that trial is testing another one of its cancer drugs, Datroway, in first-line non-small cell lung cancer. In a recent research note, though, RBC Capital Markets analyst Trung Huynh argued that AstraZeneca could still hit its revenue target even if that trial is unsuccessful.
Etcamah could still succeed elsewhere, too. A pair of ongoing trials,
CAMBRIA-1
and
CAMBRIA-2
, are testing the therapy in the “adjuvant” setting after surgery.
