Ascletis Pharma Inc. (ASCLF.PK,1672.HK) announced the initiation of a U.S. Phase I study of ASC36 oral tablets, an amylin receptor peptide agonist, for the treatment of obesity following Investigational New Drug (IND) clearance from the U.S. Food and Drug Administration (FDA).
The Phase I study will evaluate the safety, tolerability, pharmacokinetics and pharmacodynamics of ASC36 following single and multiple ascending oral doses in 86 participants with obesity or overweight.
ASC36 was discovered and developed in-house using Ascletis' Artificial Intelligence-assisted Structure-Based Drug Discovery (AISBDD), while its oral tablet formulation was developed using the company's proprietary Peptide Oral Transport ENhancement Technology (POTENT).
In non-human primate (NHP) studies, ASC36 oral tablets achieved absolute oral bioavailability of 6% to 8% at steady state, with elimination half-lives of 116 to 167 hours. The results support once-daily and potentially less frequent oral dosing.
ASC36 oral tablets reduced mean body weight by up to 13.2% from baseline after once-daily dosing for seven days in NHPs and also significantly reduced food intake.
In a head-to-head diet-induced obese rat model, ASC36 subcutaneous injection demonstrated approximately 32% and 91% greater relative body weight reduction than eloralintide and petrelintide injections, respectively, after seven days of treatment.
Ascletis said the potentially higher oral bioavailability and efficacy of ASC36 could allow for a lower oral dose and provide scalability advantages in peptide manufacturing.
The company said the ASC36 program represents its fourth Phase I peptide study initiated in 2026, further advancing its peptide pipeline and proprietary oral peptide delivery technology for obesity and other metabolic diseases.
1672.HK closed Tuesday's trade at HKD 10.1, down 1.07%.
