显示 Orphan Approval Status 为“Not FDA Approved for Orphan Indication”,与“已批准”口径冲突。

  • FDA 孤儿药数据库显示 ORZEYFUL 的 orphan designation 状态为 Designated,但 Orphan Approval Status 为“Not FDA Approved for Orphan Indication”。

Takeda (TAK) announced that the U.S. FDA has approved ORZEYFUL, an oral orexin receptor 2 (OX2R) agonist, for the treatment of narcolepsy type 1 (NT1) in adults.
Narcolepsy type 1 is a rare, chronic neurological disorder caused by orexin deficiency, affecting an estimated 120,000 people in the U.S. Patients often experience excessive daytime sleepiness, cataplexy (sudden loss of muscle tone), cognitive difficulties, and disrupted nighttime sleep, severely impacting daily life.
ORZEYFUL (oveporexton), is the first and only medicine in the U.S. designed to address the underlying cause of NT1 rather than just individual symptoms. It works by directly targeting orexin siganling, aiming to restore the pathway that is deficient in NT1 patients.
The approval is based on a comprehensive clinical program including the global Phase 3 FirstLight (TAK-861-3001) and RadiantLight (TAK-861-3002) studies. Results presented at the World sleep 2025 showed that ORZEYFUL significantly improved excessive daytime sleepiness, reduced cataplexy attacks, and enhanced overall patient function compared to placebo, with a generally manageable safety profile.
Julie Kim, President and CEO of Takeda, stated that the approval represents a new chapter for the NT1 community, introducing an entirely new class of medicine that could redefine disease management. Julie Flygare, President and CEO at Project Sleep also described the approval as a historic milestone, expanding treatment choices and offering hope for improved quality of life.
The FDA's decision was supported by Phase 3 trial data, with RT-PCR testing used to confirm cases and assess efficacy. Experts noted that ORZEYFUL enables a broader conversation in clinical practice, shifting from symptom management to disease-level treatment.
Takeda is advancing U.S. launch preparations and expects ORZEYFUL to be available following completion of the Drug Enforcement Administration (DEA) scheduling process, expecting within 90 days.
TAK has traded between $12.99 and $18.90 over the past year. The stock is closed Wednesday's trading at $16.72, up 0.97%. During the overnight trading session, the stock is at 17.16, up 2.63%.
