Oil prices slide amid hopes for progress in Mideast diplomacy
DURHAM, N.C. - Priovant Therapeutics reported Wednesday the publication of skin-specific outcomes from its Phase 3 VALOR trial of brepocitinib in adults with dermatomyositis in JAMA Dermatology, according to a press release statement.
The trial evaluated brepocitinib, an oral TYK2 and JAK1 inhibitor, in 241 subjects with dermatomyositis across 90 sites. Patients were randomized to receive brepocitinib 30 mg, brepocitinib 15 mg, or placebo.
Among patients with at least moderate itch at baseline, 54% of those receiving brepocitinib 30 mg showed clinically meaningful improvements by Week 4 compared to 10% with placebo. By Week 52, this increased to 74% versus 33%, respectively.
For patients with moderate-to-severe skin disease at baseline, 46% of those treated with brepocitinib 30 mg achieved Clear or Almost Clear skin on the Investigator’s Global Assessment by Week 52, compared to 22% with placebo. Additionally, 44% achieved functional skin remission on the Cutaneous Dermatomyositis Activity and Severity Index Activity Score, versus 21% with placebo.
Among all participants, 61.7% of brepocitinib 30 mg patients achieved clinically meaningful improvement in skin disease activity at Week 52, compared to 44.3% with placebo.
Regarding corticosteroid use, 61.7% of brepocitinib 30 mg patients receiving oral corticosteroids at baseline tapered to 2.5 mg/day or less by Week 52, compared to 34.4% receiving placebo. Some 41.7% discontinued oral corticosteroids altogether versus 23.4% with placebo.
The company reported that serious infections were increased in the brepocitinib 30 mg group compared to placebo. New or recurrent malignancy, cardiovascular events, and thromboembolic events occurred more frequently in the placebo arm than the brepocitinib 30 mg arm.
Primary efficacy and safety results from the trial were previously published in the New England Journal of Medicine.
