) announced results from its Phase 3 EXCALIBER-RRMM study showing that Zenbexus (iberdomide) in combination with daratumumab and dexamethasone demonstrated improved progression-free survival compared to a standard treatment regimen in patients with relapsed and refractory multiple myeloma.
The study enrolled 800 patients with multiple myeloma as early as first relapse. Results showed median progression-free survival of 42 months for the Zenbexus combination versus 20 months for daratumumab, bortezomib and dexamethasone, representing a hazard ratio of 0.49. The median follow-up period was 23 months.
"The results from EXCALIBER-RRMM reinforce the clinical value of Zenbexus in combination with daratumumab and dexamethasone as a treatment approach for relapsed or refractory multiple myeloma," said Cristian Massacesi, chief medical officer and head of development at Bristol Myers Squibb, according to a press release statement.
The safety profile remained consistent with previously reported findings from the study. Zenbexus received U.S. Food and Drug Administration approval under accelerated approval based on minimal residual disease-negative complete response. Continued approval may be contingent upon verification of clinical benefit in confirmatory trials.
The drug is indicated for adult patients with multiple myeloma who have received at least one prior line of therapy including a proteasome inhibitor and an immunomodulatory agent. Zenbexus carries boxed warnings for embryo-fetal toxicity and serious venous and arterial thromboembolism.
In the study, serious adverse reactions occurred in 58.3% of patients receiving Zenbexus. The most common adverse reactions included upper respiratory tract infection, fatigue, musculoskeletal pain, pneumonia and diarrhea.
