) announced today that it has submitted a supplemental New Drug Application to the U.S. Food and Drug Administration seeking to expand the approved use of Ameluz (aminolevulinic acid hydrochloride) topical gel, 10%, in combination with photodynamic therapy using the RhodoLED lamp series.
The submission seeks approval to treat mild-to-moderate actinic keratoses on the extremities, neck and trunk, extending beyond the currently approved treatment areas of the face and scalp. The application proposes treatment fields of up to approximately 240 cm² on the body with an adjusted illumination protocol. Ameluz PDT received FDA approval in September for superficial basal cell carcinoma.
The submission is based on a multicenter, randomized, double-blind, vehicle-controlled Phase 3 trial involving 172 patients with actinic keratoses on the extremities, neck and trunk, and a Phase 1 pharmacokinetics study, according to a press release statement. In the Phase 3 trial, which evaluated 4 to 15 lesions in treatment fields of approximately 80, 160 and 240 cm², 45.6% of patients treated with Ameluz PDT achieved complete clearance of all treated lesions 12 weeks after the last treatment, compared with 16.7% of patients treated with vehicle PDT.
The application includes an adjusted illumination protocol for which approval is being sought for all RhodoLED lamps and all indications. The protocol ramps light up to full intensity, holds for several minutes, then cycles on and off until the full light dose of 37 J/cm² is delivered within 15 minutes total.
Under standard FDA procedures, the agency conducts a filing review within 60 days of receipt to determine whether the application is sufficiently complete to permit substantive review.
Actinic keratosis is a pre-cancerous skin lesion caused by chronic sun exposure. In 2020, approximately 58 million people in the U.S. were affected by actinic keratosis and 13 million treatments were performed.
