S&P rises and Nasdaq jumps over 1%, rebounding after their worst week since June
PHILADELPHIA - The U.S. Food and Drug Administration approved Jemperli (dostarlimab-gxly) for adults with stage II and III microsatellite instability-high or mismatch repair deficient locally advanced rectal cancer, GSK plc (LSE/NYSE:GSK) announced today.
The approval under the accelerated pathway marks the first immunotherapy authorized for this indication. The biomarker is found in approximately 5-10% of rectal cancers.
The approval was based on data from the phase II AZUR-1 trial. Investigator-assessed data showed 68% of evaluable patients achieved sustained clinical complete responses at 12 months. An exploratory analysis indicated 76% of evaluable patients had no evidence of cancer one year or more after treatment, including 68% who reached complete response without surgery and 12 who had complete response confirmed through tissue analysis after surgery.
Independent Central Review data showed 47% of evaluable patients achieved sustained clinical complete responses at 12 months. An additional 38 participants progressing through 12-month follow-up had already achieved clinical complete response.
No disease recurrences were reported among responders after median follow-up exceeding two years, according to the company’s press release statement.
Serious adverse reactions occurred in 20% of patients receiving Jemperli. The most common adverse reactions were fatigue (32%), rash (22%), diarrhea (18%) and pruritus (17%). Treatment was permanently discontinued due to adverse reactions in 3% of patients.
The AZUR-1 trial enrolled 154 patients with previously untreated stage II and III disease. Patients received nine cycles of dostarlimab-gxly over six months, administered as 500mg intravenous infusion every three weeks.
Jemperli received FDA Fast Track and Breakthrough Therapy designations for this indication. The application received Priority Review and a National Priority Voucher, resulting in accelerated approval ahead of the PDUFA date.
Full data from the AZUR-1 interim analysis will be presented at the European Society for Medical Oncology Congress in October 2026.
