BioNTech SE (BNTX) has entered a clinical trial collaboration with Gilead Sciences (GILD) to evaluate the investigational drug Pumitamig in combination with Trodelvy (sacituzumab govitecan) as a first-line treatment for patients with advanced triple-negative breast cancer (TNBC) whose tumors have low or negative PD-L1 expression.
Triple-negative breast cancer is an aggressive form of breast cancer that accounts for about 10% to 15% of cases and is often associated with poorer outcomes that other breast cancer types. Treatment options remain limited, particularly for patients with advanced disease.
The collaboration will modify the ongoing global Phase 3 ROSETTA Breast-01 trial, which is currently evaluating pumitamig plus chemotherapy versus chemotherapy alone. Under the updated design, pending regulatory approval, chemotherapy will be replaced with Trodelvy, a TROP2-directed antibody-drug conjugate that is increasingly being used in this setting.
BioNTech will continue to sponsor and lead the study, while Gilead will supply Trodelvy for the trial.
Pumitamig is an investigational bispecific immunotherapy being jointly developed by BioNTech and Bristol Myer Squibb. The drug is designed to target both PD-L1 and VEGF-A, two pathways involved in cancer growth and immune evasion.
According to the company, ROSETTA Breast-01 will become the first global Phase 3 trial evaluating pumitamig in combination with an antibody-drug conjugate. The study follows positive Phase 2 data and reflects changes in the treatment landscape for patients with advanced PD-L1-low or PD-L1-negative triple-negative breast cancer.
Pumitamig is currently being studied in seven ongoing global Phase 3 trials across several cancer types, including lung, colorectal, gastric and breast cancers. More than 4,000 patients have received the therapy in clinical studies to date.
The combination of pumitamig and Trodelvy remains investigational, and its safety and efficacy have not been established.
BNTX has traded between $79.52 and $124.00 over the past year. The stock closed Wednesday's trading at $91.29, up 0.13%. In pre-market trading the stock is at $90.50, down 0.87%.
