AbbVie (ABBV) said Thursday its investigational drug Lutikizumab met the primary endpoint and eight ranked secondary endpoints in the Phase 3 INTREPID trial in adults and adolescents with moderate-to-severe hidradenitis suppurativa.
Hidradenitis suppurativa (HS) is a chronic inflammatory skin disease that causes painful nodules, abscesses and draining fistulas. Lutikizumab targets interleukin-1 alpha and interleukin-1 beta, which are elevated in HS lesions.
The randomized, double-blind, placebo-controlled trial assigned 1,400 patients equally to subcutaneous lutikizumab or placebo. The initial 16-week period is followed by a 36-week blinded lutikizumab period and a 156-week long-term extension.
The primary endpoint was HiSCR75 at week 16, defined as at least a 75% reduction in total abscess and inflammatory nodule count, without increases in abscess or draining fistula counts from baseline. Ranked secondary endpoints included skin pain improvement at week eight and disease flares through week 16.
At week 16, 31.4% of lutikizumab-treated patients achieved HiSCR75, compared with 23.4% receiving placebo.
At week eight, 26.2% achieved at least a three-point reduction in skin pain, versus 15% on placebo, with improvements reported as early as week one. A pain reduction of at least 30% and two points was achieved by 36% versus 22.8%, respectively.
Disease flares occurred in 18.4% of lutikizumab-treated patients during the first 16 weeks, compared with 28.1% on placebo. These comparisons were statistically significant.
Improvements were also reported on the Dermatology Life Quality Index and Hidradenitis Suppurativa Impact Assessment. IL-17A and IL-17F levels declined during the 16-week placebo-controlled period.
No new safety concerns were identified. Adverse events occurred in 53.4% of lutikizumab-treated patients and 53.7% of placebo recipients. Serious adverse events occurred in 3% versus 2.9%, while adverse events leading to discontinuation occurred in 1.7% versus 1.9%.
AbbVie expects additional results, including data from the 36-week blinded period, in 2027 to support regulatory submissions in the second half of that year. The topline findings will be presented at a future medical congress.
The company also plans to begin an HS platform study later this year evaluating lutikizumab with ABBV-295, a long-acting amylin analog, and potentially other mechanisms, including ABBV-701, an anti-TL1A antibody.
On Wednesday, ABBV closed at $284.80, down 1.48%.
Currently AbbVie shares are trading down 1.85% at $279.46.
