Ascletis Pharma Inc. (1672.HK,ASCLF.PK) announced the submission of an Investigational New Drug Application (IND) to the U.S. Food and Drug Administration (FDA) for ASC39 oral tablets, an oral small-molecule selective amylin receptor agonist, for the treatment of obesity.
ASC39 is being developed as a standalone therapy and in combination with Ascletis' once-daily oral GLP-1 receptor agonist ASC30 for the treatment of metabolic diseases, including obesity.
According to the company, in a head-to-head study in diet-induced obese (DIO) rats, once-daily oral ASC39 (5 mg/kg) for six consecutive days achieved a placebo-adjusted weight loss of 6.6%.
ASC39 demonstrated favourable pharmacokinetic profiles in rats and non-human primates, supporting its potential for once-daily oral dosing in humans.
In addition, the company said ASC39 demonstrated potency comparable to eloralintide in a head-to-head cAMP activation assay, with nearly identical EC50 values at the human amylin 1 receptor (21.4 pM vs. 21.2 pM).
"We believe amylin selectivity will be a key driver of both efficacy and tolerability in chronic weight management," said Jinzi Jason Wu, PhD, Founder, Chairman and CEO of Ascletis.
Leveraging its proprietary Artificial Intelligence-assisted Structure-Based Drug Discovery (AISBDD), Ultra-Long-Acting Platform (ULAP), and Peptide Oral Transport Enhancement Technology (POTENT) platforms, Ascletis has developed a pipeline of internally discovered drug candidates, including both small molecules and peptides.
