Genmab (GMAB) reported updated Phase 2 data showing that Petosemtamab, when combined with standard chemotherapy, continued to demonstrate encouraging response rates in patients with metastatic colorectal cancer (mCRC). The findings were presented at the ESMO Congress 2026 and support the company's ongoing global Phase 3 program.
Metastatic colorectal cancer occurs when cancer of the colon or rectum spreads to other parts of the body. While treatment options have improved, many patients continue to face poor outcomes after the disease advances.
The Phase 2 study evaluated Petosemtamab, an investigational bispecific antibody targeting EGFR and LGR5, in combination with FOLFOX or FOLFIRI chemotherapy in patients with RAS/BRAF wild-type metastatic colorectal cancer.
At median follow-up of 5.9 months in the first-line patients and 7.1 months in second-line pateitns, the therapy demonstrated confirmed and unconfirmed objective response rates of 75% in first-line patients and 67% in second-line patients. Among first-line patients with left-sided tumors, the response rate reached 81%.
The updated analysis showed that several previously unconfirmed responses were later confirmed with longer follow-up, further supporting the durability and consistency of the treatment's anti-tumor activity.
Disease control rates remained high, reaching 95% in first-line patients and 93% in second-line patients. Responses were also observed in patients with liver metastases, a group that is often associated with poorer outcomes.
The safety profile remained generally manageable. Petosemtamab-related adverse events led to treatment discontinuation in a small number of patients, and no treatment-emergent adverse events resulting in death were reported.
The updated findings provide additional support for Genmab's ongoing Phase 3 LiGeR-CRC1 and LiGeR-CRC2 trials, which are evaluating Petosemtamab in first- and second-line metastatic colorectal cancer and are currently enrolling patients.
Beyond colorectal cancer, Petosemtamab is also being studied in multiple Phase 3 trials in recurrent or metastatic head and neck cancer as Genmab continues to expand the development of the investigational therapy.
GMAB has traded between $23.62 and $44.34 over the past year. The stock closed Friday's trading (October 23, 2026) at $43.79, up 9.48%. In pre-market trading the stock is at $44.88, up 2.49%.
