On Tuesday, AstraZeneca (AZN) and Daiichi Sankyo (4568.T) said the European Commission approved Enhertu in the European Union, or EU as a standalone treatment after surgery for adults with HER2-positive breast cancer who have residual invasive disease following pre-surgical taxane-based chemotherapy and HER2-targeted treatment.
This marks Enhertu's first EU approval in early breast cancer. HER2-positive tumors produce excess amounts of a growth-promoting protein and can be aggressive.
Enhertu, or trastuzumab deruxtecan, is a HER2-directed antibody-drug conjugate jointly developed and commercialized by AstraZeneca and Daiichi Sankyo. It links a targeting antibody to a cancer-killing payload.
The approval follows a positive opinion from the European Medicines Agency's Committee for Medicinal Products for Human Use and is based on the Phase 3 DESTINY-Breast05 trial.
The randomized, open-label study enrolled 1,635 patients with residual invasive disease and a high risk of recurrence after pre-surgical treatment. It compared Enhertu at 5.4 mg/kg with trastuzumab emtansine, or T-DM1.
The primary endpoint was investigator-assessed invasive disease-free survival, measuring time to invasive cancer recurrence or death.
Enhertu reduced the risk of invasive disease recurrence or death by 53% versus T-DM1. Three-year invasive disease-free survival was 92.4%, compared with 83.7%.
The safety profile was consistent with previous findings, with no new safety concerns identified.
Based on the DESTINY-Breast05 trial, Enhertu is now approved in more than 35 countries for post-surgical treatment of adults with HER2-positive breast cancer who have residual invasive disease following pre-surgical trastuzumab, with or without pertuzumab, and taxane-based treatment.
Following the EU approval, a $75 million milestone payment is due from AstraZeneca to Daiichi Sankyo. Daiichi Sankyo recognizes Enhertu sales in most EU territories.
AZN closed on Monday at $172.06, up 0.33%. In pre-market trading, shares are down 0.35% at $171.40.
Daiichi shares are currently trading down 8.16% at JPY 2,594.50 on Tokyo Stock Exchange.
